Studies on the biogenesis of the ring systems of nicotine.

چکیده
Dawson et al. (1) have shown that the pyridine ring of nicotinic acid is utilized to form the pyridine ring of nicotine in sterile cultures of tobacco root. Since studies with animals and some microorganisms have demonstrated that tryptophan is converted to nicotinic acid (2)) tryptophan might reasonably be expected to yield the pyridine ring of nicotine. However, experiments have shown conclusively that in tobacco and some other higher plants there was essentially no conversion of C14or tritium-labeled tryptophan to compounds containing the pyridine ring (3, 4, 5). It was, therefore, necessary to examine other compounds as possible intermediates in the biosynthesis of the ring systems of nicotine. The methyl carbon of acetate, but not the carboxyl carbon, recently was shown to yield nicotine labeled significantly in the pyridine ring (6). This finding led to the postulate that in some higher plants the pyridine ring may be synthesized from simple metabolic intermediates related to acetate. The present study is an investigation of the incorporation of several W-labeled compounds, which might be interconvertible with acetate in metabolism, into nicotine in intact tobacco plants. In addition, a partial degradation of nicotine which allows for isolation of carbon from the 3 position of the pyridine ring is described.

The effect of short endurance training on the expression level of PINK-1, Parkin and PGC-1α in the heart of nicotine-sensitized rats
Background: Nicotine alters the expression of various genes in the heart. PINK-1(PTEN-induced kinase1) is the major regulator of cellular mitophagy. Moreover, Parkin is a protein that plays a key role in the process of ubiquitination. Also, PGC-1ὰ (peroxisome proliferator-activated receptor gamma coactivator 1-alpha) is the main regulator of mitochondrial biogenesis. On the other hand, exercise...
متن کاملAn In-Vitro Iontophoretic Permeation Study of Nicotine through Rat Skin
Nicotine transdermal systems are being used as an aid to smoking cessation programs. As the kinetics of nicotine delivery is important in success of a smoking cessation program, rapid and high input of nicotine is required, which is not possible by passive methods and requires enhancement strategies such as iontophoresis. Iontophoretic permeation, of nicotine looks promising, based on published...

متن کاملP-28: Effects of Resveratrol on Nicotine Toxicity of The Spermatogenesis of Rats
Background: Nicotine is a major toxic component of cigarette smoke and it is a major risk factor in the development of functional disorder of several organ systems. Resveratrol (trans-3,5,4'-trihydroxystilbene; RES) is a naturally occurring polyphenol synthesized by a variety of plant species in response to injury, UV irradiation and fungal attack. It is present in grapes, berries, peanuts...
متن کاملInvestigation of the therapeutic effect of nicotine and its metabolites on the brain
The effect of Cotinine, active metabolite of nicotine, on Aβ1-42 neurotoxicity was investigated. Cotinine possesses a longer plasma half-life, lower toxicity and it is a partial agonist of the nicotinic acetylcholine receptors (nAChR). Cotinine prolonged the survival of cortical neurons exposed to Aβ1-42. These results indicated that cotinine has a neuroprotective effect by inhibition of the fo...
